Incubation inside the bovine rumen.

چکیده

I . Details are given for the construction and use of an apparatus for the incubation of rumen liquor in semi-permeable sacs inside the bovine rumen. It was found practicable, using one animal, to incubate simultaneously samples of strained diluted rumen liquor in three identical vessels, each of 100--120 ml capacity, for periods of up to 23 h. 2. The optimal conditions for experiments of this type were investigated. It was shown that, under the conditions used, the pH value of the reaction mixtures varied little compared with the pH changes that occurred inside the rumen, and that the sampling technique was satisfactory. 3. The effects of the size and type of inoculum were also investigated. The microbial growth, measured by increases in turbidity and in concentration of protein, was greatest when the rumen liquor inoculum constituted about 20% of the reaction mixture and when the protozoa and large food particles were removed. 4. The apparatus was used to study microbial growth and hydrogenation of fatty acids in rumen liquor.

Effect of rumen bacteria from sheep adapted to a tanninferous diet on in vitro fermentation parameters of pistachio hulls using bovine inoculum

Sheep adapted to consume tannins rich feeds such as oak leaf (OL) appear to develop defensive mechanisms by their ruminal bacteria against these polyphenols. The capabilities of ruminal isolated tannins resistant bacteria from these animals to ferment a tanniniferous feed (i.e., pistachio hulls, (PH) which were incubated with rumen fluid from Holstein dairy cows was assessed. Six g positive coc...

Effects of Yucca Shidigera Extract, Probiotics, Monensin and L-Cysteine on Rumen Methanogenesis

The effects of probiotics, yucca extract, monensin and L-cysteine on rumen methanogenesis were determined by in vitro incubation using mixed bovine rumen microorganisms with ground Bermuda grass hay as a substrate. Two forms of probiotics, yucca extract, monensin and L-cysteine were tested during 24 hour anaerobic incubations. Methane and carbon dioxide production and fermentation characteristi...

Studies on Different Levels and Sources of Sulfur Supplementation on Degradative Activity of Bovine Fungi in Rumen, Using Pearl Millet Strover as Substrate (in vitro)

An extensive in vitro study was conducted to delineate the influence of levels and source of sulfur besides rumen fungi habitat on the degradation of pearl millet stover dry matter and fibrous constituents. The results revealed that degradability of dry matter, organic matter, acid detergent fibre, neutral detergent fibre and cellulose from pearl millet stover was maximized (P<0.05), irrespecti...

Characterization of the dynamics of initial bacterial colonization of nonconserved forage in the bovine rumen.

Microbial colonization is central to ruminal degradation of dietary material yet little is known about the dynamics of this process. The aim of this study was to characterize the initial stages of bacterial colonization of forage, and to assess the impact that different postsample processing and analysis methods had on the results obtained. Bacterial 16S rRNA gene-based analysis of damaged, non...

Degradation of maize protein in rumen contents. Influence of ammonia concentration.

1. The influence of ammonia concentration on the distribution of nitrogen derived from opaque-2 maize uniformly-labelled with 15N has been investigated during short-term in vitro incubation of bovine rumen contents. 2. Less 15N derived from maize was found in the non-protein-N (NPN) fraction during incubation without added NH3 than with added NH3, due entirely to differences in the amount of N ...
